Hydrogen sulfide is a novel mediator of lipopolysaccharide‐induced inflammation in the mouse

Abstract: Hydrogen sulfide (H2S) is synthesized in the body from L-cysteine by several enzymes including cystathionine-gamma-lyase (CSE). To date, there is little information about the potential role of H2S in inflammation. We have now investigated the part played by H2S in endotoxin-induced inflammation in the mouse. E. coli lipopolysaccharide (LPS) administration produced a dose (10 and 20 mg/kg ip)- and time (6 and 24 h)-dependent increase in plasma H2S concentration. LPS (10 mg/kg ip, 6 h) increased plasma H2S conce… Show more

“…H 2 S equilibrates with aqueous sulphides (HS À and S 2 À ) in biological environments and has vital roles in various physiological and pathological processes, including vasorelaxation 5 , angiogenesis 6 , cerebral microcirculation 7 , neurotransmission 8,9 , inflammation 10 , apoptosis 11 , insulin signalling inhibition 12 , oxygen sensing 13 and energy production in mitochondria 14 . Meanwhile, the H 2 S levels are related to a number of diseases, such as Down syndrome 15 , Alzheimer disease 16 , hyperglycaemia 17 , ischaemia 18 and so on.…”

“…H 2 S was administered in the form of NaHS (Sigma, St. Louis, Missouri, USA), which has been well established as a reliable donor of H 2 S (Abe and Kimura 1996;Zhao et al 2001;Deplancke and Gaskins 2003;Li et al 2005). When NaHS is dissolved in saline, about one-third of H 2 S exists as undissociated gas, and the remaining two-thirds is HS − .…”

“…In an in vivo model for myocardial ischemia-reperfusion, a U-shaped H 2 S dose dependence curve was observed, with the cardioprotective effect of H 2 S decreasing at higher concentrations (7). In a rat model of stroke, administration of high NaHS levels increased infarct volume (42), and H 2 S was found to be proinflammatory in a mouse endotoxic shock model (8). Mutations in CBS are the most common cause of severe hyperhomocysteinemia in comparison with defects elsewhere in the pathway (e.g.…”
