Hydrogel materials for the application of islet transplantation

Abstract: Type 1 diabetes mellitus is a serious disease comprising approximately 10% of all diabetes cases, and the global incidence of type 1 diabetes mellitus is steadily rising without any promise of a cure in the near future. Although islet transplantation has proven to be an effective means of treating type 1 diabetes mellitus and promoting insulin independence in patients, its widespread implementation has been severely constrained by instances of post-transplantation islet cell death, rejection, and severe advers… Show more

“…Due to the in vivo tissue stroma matrix-mimicked property, hydrogels are favorable for cell encapsulation and expansion in vitro and in vivo, enabling high-efficient tissue regeneration and cancer therapy. For instance, various cells (stem cells, 48 , 49 islet cells, 50 hepatocytes 51 , endothelial cells (ECs), 52 etc.) that were encapsulated in hydrogels could propagate and concurrently maintain functional characteristics in vitro.…”

“…A self-assembling peptide-based hydrogel is used to enhance islets functionality and survival. Although in vivo evaluation of islets encapsulated in these systems is still challenging, the in vitro evaluation showed a significant increase in their functionality and viability ( Xu et al, 2019 ).…”

“…To further investigate the in uence of highly expressed MALAT1, we transplanted β like cells transfected with lentivirus into the renal capsules of diabetic mice using alginate hydrogel (Fig. 8a, 8b) in accordance with a protocol reported in a previous study [21]. Next, we harvested β cells from the MALAT1 overexpression and NC groups about 1×10 6 before we transplanted them into STZ-diabetic mice (n = 6).…”
