Hybrid biomimetic assembly enzymes based on ZIF-8 as “intracellular scavenger” mitigating neuronal damage caused by oxidative stress

Abstract: Superoxide dismutase (SOD) was immobilized in zeolite imidazolate framework-8 (ZIF-8) through biomimetic mineralization method, namely SOD@ZIF-8, which was then used in the treatment of nerve damage by eliminating reactive oxygen species (ROS). A series of chemical characterization and enzymatic activity researches revealed that SOD was successfully embedded into ZIF-8 without apparent influence on the antioxidant activity of SOD. Cell level experiments showed that SOD@ZIF-8 could be effectively endocytosed by… Show more

“…If the PC molecules protect the cells against oxidative stress, the ROS level of the cells would decrease. As expected, for quantitative analysis using 2′, 7′-dichlorofluorescein diacetate (DCFDA) as a fluorogenic probe of ROS, , the PC-treated group showed a considerably low intracellular ROS level when compared to that of nontreated control groups (e.g., 81.8% intensity of the values in the control), indicating that the PCgels could effectively protect the neuronal cells in the hypoxia situations. The PCgels can have a vital role in regulating redox imbalances and oxidative damage in the neurodegenerative region, which is already disrupted in mitochondrial redox signaling .…”

“…These hybrid systems are not just a simple sum of their parts; they leverage the advantages of both ZIF-8 and the additional nanomaterials. 20,33,[58][59][60] A recent study has shown that ZIF-8 nanoparticles can be effectively combined with other nanomaterials for the detection and capture of specific cancer cells. This enhances the effectiveness of chemotherapy drugs and minimizes side effects.…”

“…Another study demonstrated that ZIF-8 outperformed the extensively used antibacterial ZnO ( Ahmed et al, 2019 ). The photoelectrons trapped within the zinc ions of ZIF-8 are responsible for ROS production inducing cell deformation and cytoplasm leakage of bacteria ( Ahmed et al, 2019 ; Li et al, 2022a ; Yu et al, 2022 ). It is noteworthy that the ZIF-8 crystal exhibits excellent capabilities for drug delivery because the ZIF-8 is stable in neutral or alkaline aqueous medium but break down rapidly in acidic medium ( Liu et al, 2020a ).…”
