Hybrid µCT-FMT imaging and image analysis

Abstract: Fluorescence-mediated tomography (FMT) enables longitudinal and quantitative determination of the fluorescence distribution in vivo and can be used to assess the biodistribution of novel probes and to assess disease progression using established molecular probes or reporter genes. The combination with an anatomical modality, e.g., micro computed tomography (µCT), is beneficial for image analysis and for fluorescence reconstruction. We describe a protocol for multimodal µCT-FMT imaging including the image proce… Show more

“…A diffuse phantom containing a cylindrical inclusion (100 µl), available from the FMT manufacturer, was used as previously described 4,31. The phantom was filled with diluted reference dyes in known concentrations (VM670XL at 0.40 µM, VM750 at 0.41 µM, VM790 at 0.78 µM).…”

“…Tumor vascularization was assessed with first-pass analysis of BR1 and postprocessing with MIOT (24). Functional US with the clinically approved, short-circulating BR1 microbubbles showed a tendency toward decreasing rBV values in treated and control tumors, with values being lower in B20-treated tumors (Fig 3).…”

“…One observer was blinded to the types of microbubbles and experimental groups and confirmed the results. Image analysis was done with preclinical software (Imalytics; Gremse-IT, Aachen, Germany) (24). A region of interest was defined over the whole tumor.…”

“…As the tissue detection in the technique explored in this review relies on metal-based stains, X-ray CT can also be considered as a method to detect, monitor, and even infer physiological pathways of metallic pollution in small animals . Furthermore, combination with other techniques-for instance fluorescence microscopy-may allow to identify processes within the tissues for future studies on ecophysiology and ecotoxicology (e.g., angiogenesis, apoptosis, inflammation) (Gremse et al, 2015;Handschuh, Baeumler, Schwaha, & Ruthensteiner, 2013;Metscher, 2009a). Possible detrimental effects of radiation and stain ingestion on living organisms (especially invertebrates) remain to be elucidated.…”
