Hyaluronan Receptor LYVE-1-Expressing Macrophages Maintain Arterial Tone through Hyaluronan-Mediated Regulation of Smooth Muscle Cell Collagen

Abstract: The maintenance of appropriate arterial tone is critically important for normal physiological arterial function. However, the cellular and molecular mechanisms remain poorly defined. Here, we have shown that in the mouse aorta, resident macrophages prevented arterial stiffness and collagen deposition in the steady state. Using phenotyping, transcriptional profiling, and targeted deletion of Csf1r, we have demonstrated that these macrophages-which are a feature of blood vessels invested with smooth muscle cells… Show more

“…4b) – forming two distinct PA macrophage sub-clusters. LYVE1 and SCN9A define one subset of arterial resident macrophages known to modulate arterial tone through smooth muscle collagen degradation 15,16 . The absence of these cells in the AC indicates dysfunctional arterial homeostasis in this region.…”

“…EMP-derived macrophages are characterized by high expression of Lyve-1 , Stab1, Gas6 , and Cd163 , among other genes. The hyaluronan receptor LYVE-1 has been identified as a key marker of resident macrophages in cardiovascular tissues 19 , 45 . In arteries, LYVE-1 exerts important homeostatic functions by controlling collagen expression in vascular smooth muscle cells 19 .…”

“…Thus, if tissue-resident macrophages contribute to FAP clearance, as has been reported for inflammatory, monocyte-derived macrophages (Lemos et al, 2015) some of the effects of aging or Mertk loss in macrophages might be attributed to the persistence of these cells. Finally, while the expression of Cx3cr1 in the skeletal muscle is limited to macrophages, Cx3cr1 is also expressed on monocytes and LYVE1-expressing macrophages associated with the vasculature and nerve fibers (Lim et al, 2018;Schaum et al, 2018). We therefore cannot completely exclude off target effects related to global deletion of Cx3cr1 in our system.…”
