Hyaluronan-based pericellular matrix: substrate electrostatic charges and early cell adhesion events

Abstract: Cells are surrounded by a hyaluronan-rich coat called 'pericellular matrix' (PCM), mainly constituted by hyaluronan, a long-chain linear polysaccharide which is secreted and resorbed by the cell, depending on its activity. Cell attachment to a surface is mediated by PCM before integrins and focal adhesions are involved. As hyaluronan is known to bear a negative charge at physiological pH, the relevance of its electrical properties in driving the early cell adhesion steps has been studied, exploring how PCM med… Show more

“…The influence of RGD, a simple peptide often used to enhance cell attachment, was also minimal. There are few direct studies of the influence of surface charge on cell attachment in the absence of, or without pre-treatment with, matrix ligands (Hamdan et al 2006;Fotia et al 2013). Thus, for the first time, we demonstrate that simply modifying the surface charge of a cellulose scaffold, by derivatisation using chemistry developed for the cloth dying industry, promotes attachment of cells (70% increase over unmodified cellulose scaffolds).…”

“…These results suggest that early cell adhesion is mediated by the electrostatic interaction of cationic charges with the negatively charged pericellular matrix of the cell membrane. The importance of the pericellular matrix for early cell adhesion has also been reported for other cell types such as osteoblasts, chondrocytes or epithelial cells (Cohen et al, 2003;Fotia et al, 2013;Scrimgeour et al, 2017). RGD-integrin interaction and focal adhesion formation take place in a second step when the cell-substrate distance is in a range of 25-50 nm leading again to a doubling in adherent cells compared to the cationic gel whereas the pure RGDfunctionalized gel did not show any cell adhesive properties.…”
