Hunting Interactomes of a Membrane Protein

Abstract: The identification of epitopes involved in protein-protein interactions is essential for understanding protein structure and function. Large scale efforts, although identifying the interactions, did not always yield these epitopes, could not confirm most of the known interactions, and seemed particularly unsuccessful for native intrinsic membrane proteins. We have developed a fluidics-based approach (non-steady-state kinetics) to obtain the broadest set of the epitopes interacting with a given target and appli… Show more

“…Paradoxically, hexokinase I closes VDAC, while hexokinase II seems to keep it open. The list of putative VDAC modulators is still growing – Roman and coworkers surveyed 55 novel interactions of VDAC with other proteins and small molecules (Roman et al, 2006). Furthermore, VDAC seems to mediate regulation of metabolism including calcium-dependent ATP uptake and a modification of conductance by physiological cytosolic calcium concentrations (Bathori et al, 2006).…”

“…As an essential component and regulator of mitochondrial permeability transition pore (mPTP), VDAC contributes to loss of mitochondrial membrane potential, mitochondrial swelling, and rupture of the OMM during I/R injury. Several studies have directly measured VDAC activity (Lai et al, 2006;Roman et al, 2006); however, purified VDAC in a planar bilayer might respond differently than VDAC in situ. Immunohistochemical and western blot analyses showed overexpression of VDAC1 following cerebral ischemia/reperfusion injury in rats (Li et al, 2018).…”
