Human vtRNA1-1 Levels Modulate Signaling Pathways and Regulate Apoptosis in Human Cancer Cells

Abstract: Regulatory non-protein coding RNAs perform a remarkable variety of complex biological functions. Previously, we demonstrated a role of the human non-coding vault RNA1-1 (vtRNA1-1) in inhibiting intrinsic and extrinsic apoptosis in several cancer cell lines. Yet on the molecular level, the function of the vtRNA1-1 is still not fully clear. Here, we created HeLa knock-out cell lines revealing that prolonged starvation triggers elevated levels of apoptosis in the absence of vtRNA1-1 but not in vtRNA1-3 knock-out … Show more

“…Accumulating evidence revealing a key role of the small non-coding vtRNA1-1 in conferring cellular resistance against cell death via chemoresistance 7,12 and viral defence and apoptosis 4,8,13,48 , lead us to further investigate the involvement of vtRNA1-1 in tumorigenesis and cancer progression. In agreement with its previously described function, we observed a modulation of vtRNA1-1 expression in response to apoptosis induced by physical and mechanical stress in primary human hepatocytes and liver-derived clinical biopsies (Fig.…”

“…Conversely, in Hela cells, where vtRNA1-1 was shown to confer apoptosis resistance ( Fig. 2b and 8,13 , neither changes in lysosomal pH nor in its proteolytic activity were apparent when vtRNA1-1 was depleted ( Fig. S3a, b).…”
