Human UTY(KDM6C) Is a Male-specific Nϵ-Methyl Lysyl Demethylase

Abstract: Background: UTY(KDM6C) has been reported previously to be inactive as a histone demethylase. Results: Crystallography reveals that the fold of the UTY(KDM6C) catalytic domain is highly conserved with those of KDM6A/B. UTY(KDM6C) catalyzes demethylation of Nϵ-methylated lysine histone peptides at Lys27.Conclusion: UTY(KDM6C) is a lysine demethylase that shows high structural similarity with KDM6A/B.Significance: UTY(KDM6C) is a functional Nϵ-methyl lysine demethylase.

“…While Kdm6a homozygous mutant females display severe and lethal developmental defects, males are only mildly affected (8)(9)(10)47). In light of emerging evidence of in vitro demethylase activity of KDM6C (11), the in vivo importance of KDM6A demethylase-dependent and -independent functions needs to be revisited.…”

“…The Kdm6a gene is located on the X chromosome, and conclusions on enzyme-independent functions were primarily drawn from studies with male cells containing a homolog on the Y chromosome, Kdm6c, which has no apparent enzymatic activity. However, in vitro enzymatic activity of KDM6C has been described (11).…”

“…In particular, UTX (KDM6A) is a H3K27 demethylase expressed from both X chromosomes 110 . In men, UTX has a homolog on the Y chromosome, called UTY (KDM6C), whose catalytic activity is very low or absent 111 . UTX has a tumor suppressing function and inactivating mutations in this gene are not compensated by UTY (Fig.…”

“…A paralog of the X-chromosome-located KDM6A, termed UTY (also known as KDM6C), is located on the Y-chromosome. It is so far unknown whether UTY possesses in vivo demethylase activity Walport et al, 2014). KDM6A and KDM6B take part in the regulation of gene expression programs during development (Burgold et al, 2008(Burgold et al, , 2012Lee et al, 2012;Seenundun et al, 2010).…”
