Human steroidogenic acute regulatory protein: functional activity in COS-1 cells, tissue-specific expression, and mapping of the structural gene to 8p11.2 and a pseudogene to chromosome 13.

Abstract: Steroidogenic acute regulatory protein (StAR) appears to mediate the rapid increase in pregnenolone synthesis stimulated by tropic hormones. cDNAs encoding StAR were isolated from a human adrenal cortex library.

“…Primary points of control in steroidogenesis are the transport of cholesterol from intracellular stores to the inner mitochondrial membrane, and the subsequent conversion of cholesterol to pregnenolone by the cholesterol side-chain cleavage enzyme (P450scc) [6], which initiates the synthesis of all steroid hormones. The StAR protein mediates the rate-limiting and acutely regulated step in steroidogenesis [15,29,30]. LH stimulation is mediated by the LH receptor at the first step of the signaling pathway, and subsequently intercellular cAMP increases to stimulate the transcription of the StAR in mouse MA-10 cells [36].…”

“…On the other hand, detailed studies have not yet been performed with respect to StAR expression in the CNS. Northern blot analysis revealed StAR mRNA expression in some peripheral organs (kidney, testis, ovary, and adrenal gland) but not in the brain (Sugawara et al, 1995). However, taking into account the quite low level expression of cytochrome P-450 5~mRNA, the amount of the StAR transcript expressed in the brain may also be very small.…”

“…However, defects in P450scc, adrenodoxin, and adrenodoxin reductase were eliminated by molecular genetic analysis of affected patients (5)(6)(7). Recently, the gene for steroidogenic acute regulatory protein (StAR), which functions as a labile protein factor in steroidogenesis mediating cholesterol transport within mitochondria, was cloned (8)(9)(10). Mutations in the StAR gene were subsequently identified as the cause of lipoid CAH (8,11).…”
