Human serum‐derived exosomes modulate macrophage inflammation to promote VCAM1‐mediated angiogenesis and bone regeneration

Abstract: During exogenous bone‐graft‐mediated bone defect repair, macrophage inflammation dictates angiogenesis and bone regeneration. Exosomes from different human cells have shown macrophage immunomodulation‐mediated bone regeneration potential. However, the effect of human serum‐derived exosomes (serum‐Exo) on macrophage immunomodulation‐mediated angiogenesis during bone defect repair has not been investigated yet. In this study, we explored the effects of serum‐Exo on macrophage inflammation regulation‐mediated ang… Show more

“…Furthermore, stimulating M2 macrophage phenotype by MSC-derived EVs may also improve ligament healing ( Chamberlain et al, 2021 ). Adipose tissue macrophages from lean mice release EVs that modulate macrophage polarization via contained miRNAs to promote wound healing in diabetic mice ( Xia et al, 2023 ), whereas human serum-derived EVs encouraged angiogenesis and osteogenesis by reducing the expression of M1-related genes in macrophages ( Xiang et al, 2023 ). Therapeutic EVs may also diminish the activity of M1 macrophages to alleviate periodontitis ( Luo et al, 2023 ).…”

“…When VCAM-1 is de cient, it can provoke the mobilization of neutrophils to be eliminated (Akbar et al, 2023). VCAM1 signalling in endothelial cells plays an important role in angiogenesis (Xiang et al, 2023). Atorvastatin and/or probucol lowers NF-κB levels, which resulted in the recovery of atherosclerosis in the ApoE -/mouse model (Guo et al, 2019).…”

“…VCAM1 expression was found to be overexpressed at the wound edge at 48–96 h post injury in murine excisional wounds ( Roy et al, 2008 ). The pro-angiogenic effect of VCAM1 signalling was evident in bone defect repair when treated with human serum-derived exosomes as VCAM1 positively regulates vascular endothelial growth factor (VEGF) ( Xiang et al, 2023 ). In our experimental target analysis, the reported gene silencing effect of Anti-miR-155 on FGF2, FGF7, CCL2 and VCAM1 reverted to the basal level at 72 h as the method used for introducing miRNA to the cells was based on cationic-lipid transfection.…”
