Human monoclonal antibodies derived from lymph nodes of a patient with breast carcinoma react with MuMTV polypeptides

Abstract: Nine human hybridoma cell lines were established from a fusion of axillary lymph node lymphocytes of a patient with breast ductal carcinoma with a human lymphoblastoid cell line. The human hybridoma nature of the fusion products was confirmed by chromosomal analysis and HLA typing. The hybridomas are stable over a year of growth, and can be frozen, thawed and regrown. The carcinoma cells of the patient harbor mouse mammary tumor virus (MuMTV) cross-reacting antigens. The patient's serum and the purified monocl… Show more

“…5 Subsequent laboratory work showed the ability of hormone-dependent cells to undergo repeated cycles of apoptosis secondary to cyclic hormonal withdrawal. Bruchovsky et al 6 used the androgen-dependent Shionogi carcinoma as a model to suggest that castration followed by reexposure to androgens before tumor progression preserved androgen dependence in surviving stem cells, leaving them amenable to further androgen withdrawal.…”

“…5 Detectable MMTV env gene-related antigenic reactivity was found in tissue sections of breast cancer, 6 -10 breast cancer cell lines, 11 serum, 12,13 pleural effusion 14 and lymph nodes. 15 Moreover, sequence homology to MMTV has been found in human DNA. 1,2,16 -21 Two studies detected the env gene of MMTV in 37-38.5% of breast tumor tissues but only in 0 -2% of normal tissues.…”

“…After a 5-day exposure to pokeweed mitogen (GIBCO Laboratory, Grand Island Biological Co), the lymphocytes were fused with the GM4672 human lymphoblastoid cell line according to a method described previously. 24 After fusion, the cells were seeded onto 96-well tissue culture plates (Nunc-Immunol), with hypoxanthine aminopterin thymidine selection media for 4 to 5 weeks. The clones were screened for binding to ␤2GPI with the modified aCL ELISA.…”
