Human melanoma cell lines of primary and metastatic origin express the genes encoding the chains of platelet-derived growth factor (PDGF) and produce a PDGF-like growth factor.

Abstract: Normal human melanocytes and five human melanoma cell lines were analyzed for production of plateletderived growth factor (PDGF)-like activity. Three of the melanoma cell lines released an activity that inhibited binding of 1251-labeled PDGF to human foreskin fibroblasts and stimulated [3H]thymidine incorporation in such cells. These activities were inhibited by the addition of anti-PDGF antibodies. All three factor-producing cell lines were derived from the same patient-one originated from the primary tumor (… Show more

“…In proliferation assays, the primary melanoma cell line WM-115 displayed greater sensitivity to dasatinib than the metastatic cell line WM-266-4, which is derived from the same patient [15]. The concentration of dasatinib required to inhibit 50% of the proliferation (IC 50 ) of WM-115 was of 79.3 nM (±11.7 nM).…”

“…WM-115 was derived from a vertical phase primary melanoma tumour and is dasatinibsensitive, and WM-266-4 was derived from a lymph node metastatic tumour from the same patient [15] and is dasatinib-resistant. We combined phosphoprotein enrichment with 2D-DIGE technology and mass spectrometry (MS) to investigate phospho-proteome changes caused by dasatinib treatment in the two isogenic melanoma cell lines.…”

“…There is evidence to suggest that some cell types produce only A-chain or B-chain homodimers (1-3), while human platelets contain both chains ofPDGF (4). While its exact peptide composition remains to be identified, many tissues and cell lines, mostly of mesenchymal origin, express the mRNAs encoding the A-and/or B-chains of PDGF (3,5,6). The B-chain, encoded by the c-sis protooncogene (7,8), is -60% similar in nucleotide sequence to the A-chain gene (5).…”

“…mammary carcinoma cell lines [58 -601, lung carcinoma cell lines [61], melanoma cell lines [62] and leukemia cell lines [63,641. In these cases it is highly unlikely that the endogenously produced P D G F has any autocrine function; it is possible, however, that it, in some cases, could contribute to the formation of connective tissue stroma in vivo, via a paracrine mechanism.…”
