Human mast cells produce matrix metalloproteinase 9

Abstract: Extracellular matrix-destructive enzymes, like matrix metalloproteinases (MMP), have been recognized in the process of inflammation and tissue remodeling and repair. The affected tissues often contain markedly increased numbers of mast cells. Although mast cells are capable of activating latent collagenase and proMMP, it has so far been unknown whether human mast cells themselves produce and secrete MMP9. In this study, MMP9 production by cord blood-derived cultured human mast cells and HMC-1 human mast cells … Show more

“…As most of these substances are found predominantly at the basement membrane, MMP‐9 is of particular interest in inflammatory skin diseases, where migration of cells across the basal layer to enter and leave the site of inflammation is required (31). For various cell types relevant in inflammatory skin diseases, the ability to produce MMP‐9 has been shown: epidermal keratinocytes (32), Langerhans cells (33), mast cells (34), T cells (35) and eosinophils (36). Here, we show that keratinocytes from patients with AD produce less MMP‐9 compared to keratinocytes from healthy controls.…”

“…MMP‐2/gelatinase A and MMP‐9/gelatinase B appear to perform unique defined roles during tissue injury and repair (17). Inflammatory cells, such as mast cells (18, 19), neutrophils, macrophages, lymphocytes and eosinophils, secrete MMP‐9, while a myriad of cell types release MMP‐2. Recombinant canine SCF induces mast cell MMP‐9 expression in a dose‐dependent and selective manner (18).…”

“…Mast cells have also been shown to produce MMP-9,66 and inhalation of corticosteroids reduces submucosal expression of MMP-9 in asthmatics. As interest intensifies in the potential immunomodulatory roles of MMPs, we are likely to see advances in our understanding of allergic disorders of the anterior segment, as well as presumed immuno-inflammatory disorders of the posterior segment.…”
