Human lupus‐type inclusions in umbilical cord blood mononuclear cells

Abstract: Mononuclear cells purified from umbilical cord blood activated oligoadenylate synthetase and formed human lupus-type inclusions (LI) when cultured with the purified recombinant human leukocyte interferon, IFLrA. LI frequencies increased from 0% to a low of 0.75% and a high of 6.25% in 4-day cultures with IFLrA (100 units/ml). These interferon-induced responses in the mononuclear cells of neonates indicate that LI are solely an intrinsic product of normal cells, and not an exogenous virus or some other environm… Show more

“…The unusual acid-labile IFN-␣ in sera from individuals with SLE and AIDS has an extraordinary ability to induce LI (24,30). LI are known to be products of normal cells abnormally stimulated with IFN-␣ because peripheral blood mononuclear cells from healthy adult Red Cross blood donors (31) and umbilical-cord bloods from routine births (32) form LI when cultured with IFN-␣.…”

“…These inclusions are induced both in vitro and in vivo by type I (a and /; 1) but not type 1 (y) IFN (4). The inclusions are known to be products of normal cells abnormally stimulated with a-IFN as shown by their induction in peripheral blood mononuclear cells prepared from adult Red Cross blood donors (4), and from umbilical cord bloods from routine deliveries (5).…”
