Human FAM154A (SAXO1) is a microtubule-stabilizing protein specific to cilia and related structures

Abstract: Cilia and flagella are microtubule-based organelles present at the surface of most cells, ranging from protozoa to vertebrates, in which these structures are implicated in processes from morphogenesis to cell motility. In vertebrate neurons, microtubule-associated MAP6 proteins stabilize cold-resistant microtubules through their Mn and Mc modules, and play a role in synaptic plasticity. Although centrioles, cilia and flagella have cold-stable microtubules, MAP6 proteins have not been identified in these organe… Show more

“…However, it should be noted that our approach has limitations because sometimes, as was the case with STMN2 and STMN3 in jawless fish, organisms would have closer orthologs because of the high similarity of hits or no orthologs would be found because of high dissimilarity of these genes in different taxons. As an example, MAP6 has one ortholog in Danio rerio (Dacheux et al, 2015) which our approach did not show.…”

“…By occupying greater width, a helical structure might influence the spatial organization of the microtubule network (e.g., by inhibiting microtubule co-alignment and bundling), help determine neurite or axonal width, or confer resistance to compressive loads such as might be encountered during development or regeneration (e.g., moving through a glial scar) (34). Given that MAP6 belongs to a family of four genes (MAP6, MAP6D1, FAM154A, and FAM154B) (21,35), understanding the functions of these other proteins could help clarify the special roles of various microtubule types in neurons.…”
