Human erythroid cells produced ex vivo at large scale differentiate into red blood cells in vivo

Abstract: New sources of red blood cells (RBCs) would improve the transfusion capacity of blood centers. Our objective was to generate cells for transfusion by inducing a massive proliferation of hematopoietic stem and progenitor cells, followed by terminal erythroid differentiation. We describe here a procedure for amplifying hematopoietic stem cells (HSCs) from human cord blood (CB) by the sequential application of specific combinations of growth factors in a serum-free culture medium. The procedure allowed the ex viv… Show more

“…In the second step (days 10-13), cells were diluted at 5 ϫ 10 4 or 10 5 cells/mL (for CB or mPB, respectively) and cocultured on an adherent MS5 stromal layer in basal medium supplemented with erythropoietin. In the third step (days [13][14][15][16][17][18][19][20], cells were cultured on MS5 cells in basal medium without cytokine.…”

“…16,20,21 ␥-globin genes are first expressed in fetal liver and cord blood (CB) erythropoiesis. 19,22 Shortly after birth, a switch between fetal and adult globins allows the expression of ␦-and ␤-globin genes, and the decrease of ␥-globin gene expression. 21,23,24 Adult erythropoiesis recapitulates fetal-to-adult hemoglobin switching in the so-called "compressed switch."…”

“…1 This is the concept of cultured RBC produced ex vivo from hematopoietic stem cells originating from bone marrow, peripheral blood or cord blood. 2,3 Research is already focused on the feasibility of this approach at the industrial level [US Defence Advanced Research Programs Agency (USDARPA)'s 'Blood Pharming' program (2007) (http://www.darpa.mil/dso/solicitations/baa07-21mod6.html)]. On this background, the identification of an unlimited source of stem cells is a major objective to free the production of cultured RBC from the constraints of the supply of hematopoietic stem cells.…”
