Human dendritic cells transfected with amplified MUC1 mRNA stimulate cytotoxic T lymphocyte responses against pancreatic cancer in vitro

Abstract: The CTL responses stimulated by DCs transfected with MUC1 mRNA could only recognize and lyse HLA-A2+/MUC1+ PC and other target cells under restriction by MHC class I-specific antigen presentation, providing a preclinical rationale for using MUC1 as a target structure for immunotherapeutic strategies against PC.

“…As described by Chen[ 27 ], the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) cell proliferation assay was used to defined the cell viability. In brief, RNA loaded DCs were inoculated into 96-well tissue culture plates (BD PharMingen, San Diego, CA, United States) with a density of 3000 cells per well and co-incubation with or without 3000 autologous PC cells.…”

“…Antigen-specific CTLs were produced utilizing a protocol described by Chen et al[ 2 , 26 , 27 ]. The PBMCs without adherence were cultured in serum-free medium containing 10 ng/mL IL-7 and 20 U/mL IL-2 (R&D Systems).…”

“…CD8+ T-cell responses were more effective using this combination than with the use of MUC1 cDNA alone. To target MUC1 cDNA expression only to DC, in vitro transfection of human DC with MUC1 mRNA has also been developed [46]. These MUC1-mRNA-transfected DC were able to induce MUC1-specific CD8+ T lymphocytes that can kill pancreatic cancer cells.…”

“…The unique characteristics of dendritic cells (DCs) as the most potent antigen-presenting cells (APCs) lead them to be considered as a promising tool for cancer immunotherapy [1]. In the last few years, there has been a growing interest in peptide-, RNA-, and DNA-based DC vaccines as an effective approach for cancer immunotherapy due to its promising results in achieving significance and durable treatment responses with mild adverse events [2,3]. Peptide-based vaccines provide several advantages in comparison to other types of cancer vaccines.…”
