Human Cytomegalovirus UL131-128 Genes Are Indispensable for Virus Growth in Endothelial Cells and Virus Transfer to Leukocytes

Abstract: Human cytomegalovirus (HCMV), a ubiquitous human pathogen, is the leading cause of birth defects and morbidity in immunocompromised patients and a potential trigger for vascular disease. HCMV replicates in vascular endothelial cells and drives leukocyte-mediated viral dissemination through close endothelium-leukocyte interaction. However, the genetic basis of HCMV growth in endothelial cells and transfer to leukocytes is unknown. We show here that the UL131-128 gene locus of HCMV is indispensable for both prod… Show more

“…Because the ULb= genes have been retained through the evolution of HCMV and are conserved in all clinical strains, these genes are hypothesized to play important roles in viral persistence in the host, functioning in immune evasion, replication, or latency in cell types other than fibroblasts. Consistent with this hypothesis, viruses lacking the ULb= region are altered in their ability to cause disease (22), to establish/maintain latency (23,24), and to infect clinically relevant cells, including endothelial cells, epithelial cells, and leukocytes (22,25,26).…”

“…In general, mutants that lack gH/gL/UL128-131 replicate as well as or better than wild-type strains on fibroblasts (21,(27)(28)(29). This indicates that gB-mediated fusion with fibroblast membranes can be facilitated by gH/gL/gO.…”

“…The gH/gL/gO complex appears to be sufficient for HCMV entry and replication into fibroblasts (23,24) but still is required for proper growth and for fusion events in all cell types (15,16,25). The more recently discovered gH/gL/ UL128/UL130/UL131A complex (Pentamer) instead is required for endothelial/epithelial cell, leukocyte, and monocyte tropism (26)(27)(28). On the cellular side, several putative HCMV receptors have been described, but none of them completely fulfills the characteristics of an entry receptor (17,29).…”
