Human Corneal Endothelial Cells Expanded In Vitro Are a Powerful Resource for Tissue Engineering

Abstract: Human corneal endothelial cells have two major functions: barrier function mediated by proteins such as ZO-1 and pump function mediated by Na-K-ATPase which help to maintain visual function. However, human corneal endothelial cells are notorious for their limited proliferative capability in vivo and are therefore prone to corneal endothelial dysfunction that eventually may lead to blindness. At present, the only method to cure corneal endothelial dysfunction is by transplantation of a cadaver donor cornea with… Show more

“…However, creating an endothelial sheet (ES) might be the most important challenge to overcome. Unlike murine, rabbit, and bovine cells, human CECs have limited proliferative capability in vivo . Although decellularized and layered human corneal lamellae have shown biocompatibility with human CECs, making endothelialized human lamellae is a feasible option for endothelial keratoplasty in the future.…”

Currently, re-endothelialization by human CECs is only possible in vitro.…”

“…Despite the known importance of Collagen IV, it remains unclear of the mechanism of action in how it promotes cell attachment and growth on atelocollagen sheets. It also remains unclear how collagen IV may affect the behavior of HCEC aggregates (not single cells) such as phenotype on plastics [10][11][12][13][14][15][16][17][18] on atelocollagen sheets.…”

“…It plays a significant role in maintaining visual function: controls cornea hydration and transfers nutrients and other molecules from the aqueous humour [9]. As a result of aging, diseases, injury, or surgeries EC could be damaged and influence corneal blindness [10]. The minimum density of corneal EC for normal functioning is thought to be around 500 cells/mm [11].…”
