Human Colon Cancer–Derived Clostridioides difficile Strains Drive Colonic Tumorigenesis in Mice

Abstract: Defining the complex role of the microbiome in colorectal cancer and the discovery of novel, protumorigenic microbes are areas of active investigation. In the present study, culturing and reassociation experiments revealed that toxigenic strains of Clostridioides difficile drove the tumorigenic phenotype of a subset of colorectal cancer patient–derived mucosal slurries in germ-free ApcMin/+ mice. Tumorigenesis was dependent on the C. difficile toxin TcdB and was associated with induction of Wnt signaling, reac… Show more

“…The gut microbiota is closely related to CRC. In addition to Fusobacterium nucleatum , Enterotoxigenic Bacteroides fragilis , and PKS + Escherichia coli , which have been confirmed earlier [ 5 7 ], in recent years, various bacteria, such as Porphyromonas gingivalis , Peptostreptococcus anaerobius , and Clostridium difficile have been confirmed to be associated with CRC [ 8 10 ].…”

“…These data along with permeability data suggest that there is a concerted response to maintain barrier function in terms of permeability during infection that may not be present in homogenous cell culture models and explant intoxication models. Indeed, studies have reported increased crypt length and epithelial cell differentiation during infection, which may reinforce the mucosal barrier during CDI 65, 66 . Chemical inhibition of ENaC revealed that there was no difference in function during infection.…”

“…For example, metagenomics is combined with metabolomics ( Oh et al, 2020 ). Meta-analysis attempts to systematically merge data across multiple studies and transform it into metadata that can be analyzed simultaneously ( Armour et al, 2019 ; Wang et al, 2021 ; Drewes et al, 2022 ), which reduce study bias, increase statistical power and improve overall biological understanding of a study effect. As for data mining, machine learning has been applied to find biomarkers and carry out classification or prediction tasks, such as diagnosis, disease course, and disease severity ( Marcos-Zambrano et al, 2021 ).…”
