Human Breast Tumor Cells Express Multimodal Imaging Reporter Genes

Abstract: This study confirmed the usefulness of eGFP-tk in many applications by providing, in vitro and in vivo, the sensitive and reporter gene-specific imaging. ZR75-1-eGFP-tk cells that are ready to incorporate in various imaging platforms constitute a useful model in breast cancer research.

“…While the first descriptions for this reporter construct were prior to the 2005 cutoff for this review,299,300 they are still actively employed in ongoing investigations particularly for labeling and observation of tumor cells,301,302 and they have been used to monitor promoter activity 303. However, fusion reporters are not without their limitations: it is quite common for a subunit in a fusion product to be inferior in performance to the freestanding protein.…”

“…Imaging of transcriptional regulation of endogenous genes provides information on the many cellular processes associated with cancer [73], allowing the determination of gene profiles of different tumours [10,74]. This helps to improve the diagnosis of different types of tumours such as breast cancer [36,43].…”

“…PET/CT or SPECT/CT systems could both be utilized to image the HSV-tk metabolite (Figure 4a). 47 In other example, Kang et al has described a trimodality approach for sentinel lymphocyte detection, whereby OI was utilized for initial detection of signal, after which the signal from PET imaging was coregistered with both MRI (Figure 4e) and CT (Figure 4b) images. 48…”
