Human Breast Fibroblasts Inhibit Growth of the MCF10AT Xenograft Model of Proliferative Breast Disease

Abstract: Stromal fibroblasts are important for normal breast homeostasis and regulation of epithelial growth; however, this regulatory function is altered during carcinogenesis. To study the role of fibroblasts in the development of breast cancer, fibroblasts derived from normal breast (NAFs) were incorporated into the MCF10AT xenograft model of progressive proliferative breast disease. The persistence of human NAFs in xenografts was established by intracellular labeling and tyramide-coupled fluorescent in situ hybridi… Show more

“…Cells were incubated with a rabbit polyclonal antibody to Ki-67 (dilution 1:50, Zymed/Invitrogen) for 1 h. Secondary detection was accomplished using a streptavidin–biotin system, as previously described [21]. The percentage of Ki-67 labeled cells was determined after counting of cells in random, 200× microscopic fields.…”

“…Tissues were pretreated with 3% H 2 O 2 and denatured in 70% formamide in 2ϫ SSC (1ϫ SSC is 0.15 M NaCl plus 0.015 M sodium citrate) (pH 7.0, 74°C, 2 min). The remaining procedures essentially matched those previously described (21), which were modified from the Roche Nonradioactive In Situ Hybridization Application Manual. Briefly, biotinylated probe was denatured at 74°C for 10 min and applied to mammary sections for overnight hybridization at 37°C.…”

“…A number of reports have also demonstrated fibroblast effects on growth differentiation and invasion of normal/malignant breast epithelial cells 9, 27–34. The importance of the functional state of the stromal cell compartment for the clinical behavior of tumors was recently reported 35…”
