Human antibody response to lipopolysaccharides fromNeisseria gonorrhoeae

Abstract: (Cohen, Kellogg, and Norins, 1969). It must, of course, be remembered that in these experiments the inoculum, and hence the antigenic stimulus, was very large, certainly more than would be found in the early days of a new natural infection. The traditional gonococcal complement-fixation test (Price, 1930) is used in the diagnosis of chronic rather than acute cases and even then is not regarded as very reliable. In recent years several new tests have been described though none are yet in routine use.Cohen and hi… Show more

“…Buchanan, Swanson, Holmes, Kraus, and Gotschlich Our observation that more females than males infected with gonococci have elevated antibody levels to gonococcal pili is consistent with nearly every previous report of serologic tests for gonorrhea (27,30,40,(43)(44)(45)(46)48) and may be related to the duration of infection in each sex. Most men develop dysuria and urethral discharge within 5 days after infection and seek diagnosis or treatment.…”

The use of a pure antigen preparation decreases the chance of detecting crossreacting antibody responses to organisms other than gonococci, and is a prerequisite to quantitating the antibody response to a single antigen.…”

“…Killing of serum-sensitive (unsiaylated) gonococci by non-immune normal human sera is initiated at least in part by IgM directed against LOS (Ward and Glynn 1972;Schneider et al 1982;Rice 1989;Griffiss et al 1991;Jarvis 1995); killing by non-immune sera is reduced by LOS sialylation (Parsons et al 1989;de la Paz, Cooke and Heckels 1995). LOS sialylation reduced binding of some (Elkins et al 1992) but not all (Wetzler et al 1992;de la Paz, Cooke and Heckels et al 1995) antibodies directed against PorB.…”

“…These results indicate that, like other attempts to identify antibodies against gonococcal LPS 1972;Watt, Ward and Glynn, 1971;Maeland and Matre, 1975), screening of sera for bactericidal activity against LPS-group-I strains will not be of use as a diagnostic test.…”
