HscA and HscB Stimulate [2Fe-2S] Cluster Transfer from IscU to Apoferredoxin in an ATP-Dependent Reaction

Abstract: The role of the Azotobacter vinelandii HscA/HscB co-chaperone system in ISC-mediated iron-sulfur cluster biogenesis has been investigated in vitro by using CD and EPR spectrometry to monitor the effect of HscA, HscB, MgATP, and MgADP on the time course of cluster transfer from [2Fe-2S] IscU to apo-Isc ferredoxin. CD spectra indicate that both HscB and HscA interact with [2Fe-2S] IscU and the rate of cluster transfer was stimulated more than 20-fold in the presence stoichiometric HscA and HscB and excess MgATP.… Show more

“…The presence of NTPase enzymes in the assembly of metal clusters appears to be an emerging theme in metallobiochemistry, although definitive roles have not yet been established in many cases (28)(29)(30)(31)(32)(33)(34). HydF contains an N-terminal GTPase domain comprised of the Walker A P-loop and Walker B Mg 2þ binding motifs (8), as well as a C-terminal domain containing the conserved amino acids CXH X44-53 HCXXC comprising the putative ligands for an iron-sulfur cluster (7).…”

“…The apo form of ferredoxin (aFd), has classically served as a [2Fe-2S] cluster acceptor protein in different biochemical studies (Wu et al, 2002;Wu and Cowan, 2003;Chandramouli and Johnson, 2006). We therefore used At-NEET as a potential [2Fe-2S] cluster donor and aFd as a potential [2Fe-2S] cluster acceptor ( Figure 7) to test the hypothesis that NEET could act as an Fe-S transfer protein.…”

“…This motif is missing in SufU-type and NifU-type scaffolds, which are not usually encoded with HscA or HscB, suggesting that SufU and NifU either do not require molecular chaperones or utilize a completely separate chaperone system (46). Recent work has shown that A. vinelandii HscA and HscB can enhance transfer of [2Fe-2S] clusters from IscU to apoproteins in an ATP-dependent manner (20). Deletion of hscA or hscB does diminish the activity of succinate dehydrogenase (SDH or complex II) in E. coli (121).…”
