HPLC–UV method for temozolomide determination in complex biological matrices: Application for in vitro, ex vivo and in vivo studies

Abstract: A high-performance liquid chromatography method for temozolomide (TMZ) determination in complex biological matrices was developed and validated for application in in vitro, ex vivo and in vivo studies of new nanotechnology-based systems for TMZ nasal delivery. The method was able to quantify TMZ in nanoemulsions, following cellular uptake, in the porcine nasal mucosa and in mouse plasma and brain. Analyses were performed on a C 18 column at 35°C, under UV detection at 330 nm. The mobile phase was methanol-acet… Show more

“…Intracellular TMZ levels were quantified using high-performance liquid chromatography (HPLC) according to protocols from previous studies with modifications. 20 GPCs were treated with p38MAPK inhibitors for 24 h, followed by 500 µM TMZ. The cells were collected and lysed at 0, 5, 15, 30, 60, 120, and 180 min after TMZ exposure using modified RIPA lysis buffer (pH 3) containing 100 µM dexamethasone as the internal standard.…”

“…Blank NP was also produced for comparative purposes. The encapsulation efficiency analyses (EE%) of TMZ into PLGA nanoparticles were performed applying validated reverse-phase high-performance liquid chromatography (RP-HPLC) methodology adapted from Michels and co-workers ( Michels et al, 2019 ) with standard analytical curve built-in acetic acid 0.5% (y = 74177x + 1360,6; r 2 = 1,00).…”

“…Standard solutions of TMZ have been prepared in the 10–180 µg/mL concentration range by appositely diluting in water a stock TMZ solution containing 147.1 mg of TMZ in 25 mL of a 1:1 mixture of MeOH and diluted aqueous HCl (pH = 4.5). After microfiltration of the prepared solutions, they have been injected in HPLC by adapting a method described previously 52 . In particular, HPLC analysis was performed on an Agilent LC1120 system equipped with a manual injector using a C-18 column (300 mm × 4.6 mm i.d., 5 μm particle size) and a methanol/acetic acid (0.03% in water) 35/65 as mobile phase.…”
