HPLC Determination of Guanidino Compounds in Serum of Uremic Patients Using Pyridoin as Derivatizing Reagent

Abstract: & Guanidino compounds: guanidine (GD), methylguanidine (MG), guanidinoacetic acid (GAA), guanidinopropionic acid (GPA), guanidinobutyric acid (GBA), arginine (Arg), guanidinosuccenic acid (GSA), and creatinine (CRN) as derivatives of pyridoin were separated from Kromasil C-18, 5 lm column, when eluted with methanol-water-sodium tetraborate buffer (0.1 M, pH 8.8) (57:28:15 v=v=v) with a flow rate of 1 mL=min. The UV detection was at 228 nm, linear calibrations were observed with 1.14-141 lmol=L and limits of de… Show more

“…A number of derivatizing reagents have been proposed for HPLC determination of guanidino compounds, mainly benzoin, [12][13][14] ninhydrin, 15,16 9,10-phenanthrenequinone, 17 anisoin, 18 furoin 19 and pyridoin. 20 Gas chromatography has high potential for the separation of organic compounds with low running cost, and analysis could be completed with short running time. The GC of guanidino compounds has been carried out aer precolumn derivatization with triuoroacetic anhydride, 21 hexauoroacetylacetone, 22 hexa-uoroacetylacetone mono (trimethylsilyl) triuoroacetamide, 23,24 hexauoroacetylacetone pentauorobenzyl bromide, 25 hexa-uoroacetylacetone and ethyl chloroformate.…”

“…The hydrogen atom of the amino function may be substituted [1]. These are basic compounds and many of these are present in biological fluids and in tissues at low concentration in human and animals [2]. Metformin (N,Ndimethylguanidine) ( Figure 1) is used as an oral antihyperglycemic drug in the management of non-insulin dependent diabetes mellitus [3,4].…”

“…The analytical procedures are mainly based on high performance liquid chromatography (HPLC), 5,[9][10][11][12][13][14][15][16] gas chromatography (GC) 6,[17][18][19][20] and capillary electrophoresis. [21][22] HPLC is more frequently used for the analysis of guanidino compounds, 4 but capillary GC has high potential for the separation and analysis of volatile organic compounds with short analysis time.…”
