HPLC Analysis of Allicin and Other Thiosulfinates in Garlic Clove Homogenates

Abstract: Reversed-phase high Performance liquid chromatography (C18-HPLC) was used to separate and quantitate all the detectable alkyl and alkenyl thiosulfinates, including configurational isomers, of garlic homogenates. Pure thiosulfinates were synthesized or isolated and identified by (1)H-NMR, and their extinction coefficients determined. Some configurational isomers required Separation by silica-HPLC. Five previously unreported thiosulfinates have been found, four of which contain the TRANS-1-propenyl group and inc… Show more

“…(C. albicans, C. glabrata, C. parapsilosis, C. tropicalis and C. crusei. 7 In spite of geographic variation, 'garlic extract liquid' for our isolates are consistent with those of Sivam, 8 but are relatively lower than values obtained in the literature. 9 This antimicrobial potency disparity of garlic has been attributed to the different concentrations of individually and synergistically active biosubstances in garlic preparations coupled with their interactions with sulfhydryl agents in culture media.…”

“…(2-propenyl E-1-propenethiosulfinate; 2P1PTS) and 2-propene-1-sulfinothioic acid S-(E,Z)-1-propenyl ester ((E,Z)-1-propenyl 2-propenethiosulfinate; 1P2PTS) by comparison of the obtained NMR and MS data with that previously reported (Lawson et al, 1991;Block et al, 1996;Calvey et al, 1997;Yoshida et al, 1999). Both compounds are known to be present in garlic homogenates (Lawson et al, 1991;Block et al, 1992).…”

“…Reportedly, in unheated garlic puree the concentrations of 2P1PTS and 1P2PTS are almost equal (Lawson et al, 1991). The reason for the synergistic effect observed in color developing activity, (0.5-2P1PTS + 0.5-1P2PTS) > 2P1PTS > 1P2PTS, has not yet been elucidated.…”

“…Allicin was isolated from garlic extract using preparative silica gel thin layer chromatography using toluene:ethyl acetate (100:30) as developing solvent then identified by spectroscopic data. The resulting spectra were compared with reported data (Cruz-Villalon, 2001;Lawson et al, 1991;Jansen et al, 1987). Pure compound was used to prepare calibration curve.…”
