How Does Calcium-Dependent Intracellular Regulation of Voltage-Dependent Sodium Current Increase the Sensitivity to the Oxadiazine Insecticide Indoxacarb Metabolite Decarbomethoxylated JW062 (DCJW) in Insect Pacemaker Neurons?

Abstract: Decarbomethoxylated JW062 (DCJW), the active component of the oxadiazine insecticide (S)-methyl 7-chloro-2,5-dihydro-2-[[(methoxycarbonyl)[4-(trifluoromethoxy)phenyl] amino]carbonyl] indeno[1,2-e][1,3,4]oxadiazine-4a(3H)-carboxylate (DPX-JW062)(indoxacarb), was tested on 2 inward voltage-dependent sodium currents (named INa1 and INa2) expressed in shortterm cultured dorsal unpaired median neurons of the cockroach Periplaneta americana. Under whole-cell voltageclamp conditions, application of DCJW resulted in a… Show more

“…Cyclosporine A (CsA) is an effective immunosuppressant, a calmodulin inhibitor, restraining the production of interlukin-2 and other cytokine by inhibiting calcineurin (3,4) and by accordingly refraining the activity of T-cell. The medicine was discovered in 1969-1970 and has a unique immunosuppressive activity.…”

“…Because they express membrane receptors and ion channels targeted by insecticides, they constitute ideal biological models to investigate the mode of action of insecticides. More recently, the use of such biological models have highlighted that intracellular regulatory mechanisms have fundamental consequences for the sensitivity of insecticide targets Lavialle-Defaix et al, 2010). However, among insect neuronal preparations available, the fully differentiated Mosquito Isolated Neurons (MIN) model is still lacking.…”

“…Tissue expression pattern of PaTBH transcripts was performed by RT-PCR experiments using the sense primer GSP1 and the reverse primer GSP3 (Table 1) as described previously (Moignot et al 2009). For multiplex single-cell RT-PCR, isolated dorsal unpaired median (DUM) neurons were prepared as already described (Lavialle-Defaix et al 2010). The cytoplasm of a single neuron was harvested via a patch-pipette mounted on a micromanipulator.…”
