How best to halt and/or revert UV‐induced skin ageing: strategies, facts and fiction

Abstract: Once considered mainly a cosmetic issue, photoageing research has long moved to the forefront of investigative dermatology. Besides obvious market pressures, increasing insight into the mechanistic overlap between UV-induced skin cancer and UV-induced skin ageing has contributed to this development. Also, as strategies that work to antagonize intrinsic skin ageing/senescence may also be exploited against photoageing (and vice versa!), it has become an important skin research challenge to dissect both the diffe… Show more

“…Briefly, 6 cm culture dishes of 80% confluent HEK 293T cells were switched to antibiotic-free media 24 h before transfection. HEK 293T cells were transfected using TransIT-2020 transfection reagent (Mirus Bio LLC, Madison, WI) with pcDNA-OA1-GFP, VSV-G, and pCMVdR8.91 plasmids [68] at a 3∶2:1 ratio. 3 h after addition of transfection mix, cells were switched to high BSA media.…”

“…The first is intrinsic or innate aging, an unpreventable process, which affects the skin in the same pattern as it affects all internal organs. The second is extrinsic aging, which is the result of exposure to external factors, mainly ultraviolet (UV) irradiation, that is also referred to as photoaging 1 . Intrinsic skin aging is influenced by hormonal changes that occur with age, 2 such as the gradual decreased production of sex hormones from the mid-twenties and the diminution of estrogens and progesterone associated with menopause.…”

“…Among other things, skin aging is strongly associated with the loss of skin moisture. 2 , 4 , 8 The HA content in the dermis is significantly higher than that in the epidermis, and the papillary dermis has much higher levels of HA than the reticular dermis. 13 The most dramatic histochemical change observed in senescent skin is the marked disappearance of epidermal HA, and a progressive reduction in the size of the HA polymers in the skin as a result of aging has been reported.…”

“…1 Ground substance is a highly hydrophilic, viscous, and complex constituent such that the exchange of nutrients and metabolic waste between cells, fibers, and blood supply is best facilitated by the hydrated nature of the ECM. 1 Skin aging is caused by intrinsic and extrinsic aging 2 , 3 and also by a loss of skin moisture, 4 which can be considered a major causative factor.…”
