Host Immune System Gene Targeting by a Viral miRNA

Abstract: Virally encoded microRNAs (miRNAs) have recently been discovered in herpesviruses. However, their biological roles are mostly unknown. We developed an algorithm for the prediction of miRNA targets and applied it to human cytomegalovirus miRNAs, resulting in the identification of the major histocompatibility complex class I-related chain B (MICB) gene as a top candidate target of hcmv-miR-UL112. MICB is a stress-induced ligand of the natural killer (NK) cell activating receptor NKG2D and is critical for the NK … Show more

“…Previous reports indicated that some viruses modulate host immune and apoptosis-related pathways, mostly as a kind of immunoevasion strategy [1,3,8]. Our statistical npg findings were consistent with these reports, while suggesting additional new signaling pathways.…”

“…Cellular targets of HCMV-encoded miRNAs include gene products with functions relating to control of cell cycle, secretory cellular pathways and, of particular interest, immune evasion. [54][55][56] Most of these targets have been analysed during lytic infection and, again, are outside the scope of this article but have recently been reviewed. 50 However, we will discuss the role of viral miRNAs in targeting gene expression associated with immune evasion mechanisms below.…”

“…Two likely scenarios for viral miRNA function include targeting of cellular gene expression to induce a more favorable environment for the virus, or the regulation of viral genes to establish precise temporal or tissue specific regulation of viral gene expression. Two recent studies would suggest that both scenarios occur and that a single viral miRNA is capable of regulating both cellular and viral transcripts (Grey et al, 2007;Stern-Ginossar et al, 2007).…”
