Host-Guest Chemistry in Supramolecular Theranostics

Abstract: Macrocyclic hosts, such as cyclodextrins, calixarenes, cucurbiturils, and pillararenes, exhibit unparalleled advantages in disease diagnosis and therapy over the past years by fully taking advantage of their host-guest molecular recognitions. The dynamic nature of the non-covalent interactions and selective host-guest complexation endow the resultant nanomaterials with intriguing properties, holding promising potentials in theranostic fields. Interestingly, the differences in microenvironment between the abnor… Show more

“…Inclusion of the toluene molecule does not affect the crystal parameters of ε-phase, and the framework arrangement, identical to that observed in experiments using DCM, is maintained (see Table 1). Guest molecules mainly interact with the host frameworks through noncovalent binding [28]. Intermolecular interactions of toluene and γ-CD molecules in ε•tol-phase were quantified using Hirshfeld surface analysis.…”

“…The host generally has a cavity-like space or a suitable space to hold another molecule, which is then called the guest. Such systems are also recognised in biology [3], where they are denoted as enzyme-receptor systems and/or lock-key system. Protein folding processes involve excellent examples of host-guest systems in biology [4].…”

“…Currently, a main driving force for the interest in host-guest systems is their potential role in cutting-edge advances in fields such Ogoshi et al in 2008 [19]. The progress in host-guest chemistry of pillar [n]arenes is well documented in a recent review by Guo et al [3].…”

“…During the past decades, numerous supramolecular host molecules have been developed as artificial receptors, and their host-guest interactions as well as their derived supramolecular materials have shown significant potentials in biomedical applications 25-31. Based on host-guest interactions, supramolecular host molecules may influence the physiochemical and biological properties of guest species, such as solubility, stability, toxicity and therapeutic efficacy 32-35. As one of the most prominent example in this area, Sugammadex, a γ-cyclodextrin derivative, has been approved by more than 50 countries, including the European countries and the U.S.A., as a specific reversal agent for some of the non-depolarizing steroidal NMBAs including rocuronium and vecuronium 36, 37.…”
