Host-based lipid inflammation drives pathogenesis in Francisella infection

Abstract: Mass spectrometry imaging (MSI) was used to elucidate host lipids involved in the inflammatory signaling pathway generated at the host-pathogen interface during a septic bacterial infection. Using as a model organism, a bacterial lipid virulence factor (endotoxin) was imaged and identified along with host phospholipids involved in the splenic response in murine tissues. Here, we demonstrate detection and distribution of endotoxin in a lethal murine infection model, in addition to determining the temporally and… Show more

“…The dual-polarity lipid imaging matrix, norharmane (NRM) 21,27 was prepared at 7 mg/mL in a solution of 2:1 chloroform:methanol (v:v). The NRM solution was applied using an HTX TM-sprayer (HTX Technologies, Chapel Hill, NC) using the following settings: 30 °C nozzle temperature, 10 passes, 0.120 mL/min flow rate, 1200 mm/min velocity, 3 mm track spacing, 10 psi pressure, 3 L/min gas flow, 30 s dry time between passes, using the “CC” coverage pattern.…”

“…Matrix-assisted laser desorption/ionisation-mass spectrometry imaging (MALDI-MSI) is a powerful method for visualising the spatial distributions of lipids throughout biological tissues [1][2][3]. Its versatility for mapping changing lipid compositions within tissues has been demonstrated in a variety of applications, including oncology [4][5][6], bacterial infections [7,8] and liver disease [9,10], amongst many others. The heterogeneous lipid compositions observed with MSI are ultimately the result of cellular-level lipid metabolism occurring within the cells constituting the tissue sample.…”

“…As part of the host response against infection, lipid responses receive increasing attention in the scientific community ( 51 ). Little is known about how exogenous PEA can modulate other lipid mediators involved in neuroinflammation.…”

“…No data are available during infection. Here, we used targeted quantification of lipids to evaluate whether PEA could modify the levels of several bioactive lipids ( 51 ). In the systemic circulation, prophylactic PEA significantly reduced concentrations of two pro-inflammatory mediators: AA and one of its metabolites, 20-HETE which unveils a novel anti-inflammatory mode of action of PEA during infection.…”
