Hormone and carbohydrate metabolism associated genes play important roles in rhizome bud full‐year germination of Cephalostachyum pingbianense

Abstract: Cephalostachyum pingbianense is the only woody bamboo species that can produce bamboo shoots in four seasons under natural conditions. So far, the regulatory mechanism of shoot bud differentiation and development is unknown. In the present study, indole-3-acetic acid (IAA), zeatin riboside (ZR), gibberellin A3 (GA 3 ) and abscisic acid (ABA) contents determination, RNA sequencing and differentially expressed gene analysis were performed on dormant rhizome bud (DR), growing rhizome bud (GR), and germinative bud… Show more

“…The declining IAA content was accompanied by an accumulation of IAA-Asp, confirming a low ratio of IAA to IAA-Asp in the axillary buds after decapitation. The germination of axillary buds after decapitation was closely related to the decreasing synthesis of ABA, which is consistent with the study conducted using woody bamboos [ 29 ]. In addition, ABA showed a continuous downward trend after decapitation, implying that perhaps ABA not only plays a role in the germination of axillary buds but also in the subsequent bud growth.…”

“…Horseradish peroxidase enzyme-catalyzed label-antibody complexes were formed by combining antibodies directed against GA, IAA, ABA, ACO and ACS with enzyme-catalyzed label and hormones, which generates a blue material when combined with TMB substrate solution. Spectrophotometric measurements were then performed at 450 nm (Infinite M200 pro, Tecan, Switzerland) (Gao et al, 2022;Li et al, 2022). In the Excel worksheet, the linear regression curve was plotted using the standard concentration as the horizontal coordinate and the corresponding OD value as the vertical coordinate, and the concentration value of each sample was calculated according to the curve equation.…”

“…The rhizome bud germination of bamboo, i.e., bamboo shooting, is the main pathway of asexual reproduction in woody bamboos, as well as the foundation of bamboo resource cultivation and bamboo industry [14][15][16]. According to the type of underground rhizome, woody bamboos can be totally divided into two types: the scattered bamboos and the clump bamboos, which display different shooting phenologies.…”

“…On the other hand, the clump bamboos occur in tropical regions (such as Dendrocalamus latiflorus) and have a shoot-sprouting period from June to August. Both types start shooting soon after the beginning of the rainy season [15,16]. Therefore, empirically, the rhizome buds germination of woody bamboos with a single shooting peak is generally considered to be induced by the regulation of endogenous hormone synthesis and transport following an increase in soil temperature and precipitation in the rainy season [8].…”
