This review addresses direct and indirect metabolic actions of hormones co-encoded in the preproglucagon gene of fishes. Emphasis is placed on a critical analysis of the effects of glucagon and glucagon-like peptide (GLP) and the current knowledge of the respective modes of action is reviewed. In mammals GLPs exert no direct metabolic actions. In fish liver, GLP and glucagon act on similar targets of intermediary metabolism by enhancing flux through glycogenolysis, lipolysis and gluconeogenesis. Increases in substrate oxidation are not uniform. Hormonal activation of glycogen phosphorylase and triglyceride lipase and inhibition of pyruvate kinase are implicated in these actions. Hormone-dependent hyperglycemia, depletion of hepatic glycogen and increases in free fatty acids are noticeablein vivo. Glucagon also activates hepatic amino acid uptake and ammonia excretion.Glucagon actions are accompanied by large increases in hepatic cAMP and increased phosphorylation of pyruvate kinase. Metabolic effects measured after GLP administration are associated with minor, if any, increases in cAMP and effects on pyruvate kinase are variable. We hypothesize that different hepatic receptors with differing modes of intracellular message transduction are involved in glucagon and GLP actions while targetting identical metabolic routes. Responses of different species of fish cover a wide spectrum, and variation of response with the circannual cycle of experimental animals makes comparisons of results, even within one species, difficult.

ABSTRACT:Various endocrine and physiological responses of fish exposed to forceful physical and chemical stimuli are reviewed with emphasis on the effects of crude oils and their hydrocarbon constituents. The chemistry and toxicity of petroleum hydrocarbons are examined and methods for experimental exposure of fish to crude oil and petroleum hydrocarbons are considered. A variety of blood-borne parameters recognized as reliable tools in determining the relative severity of stress in fish are reviewed. The effects of stress and petroleum hydrocarbons on endocrine responses including changes in plasma AL-KINDI, BROWN, and WARING catecholamines, corticosteroids, and thyroid hormones are reviewed. The physiological responses: changes in plasma glucose, osmotic and ionic regulation, blood oxygen, hematocrit and hemoglobin concentration are explored, and histopathological effects of crude oil on fish are reviewed. Recent studies of the effects of petroleum hydrocarbons on fish larvae are considered and the increased sensitivity of the early life stages of fish are highlighted.

The present study was aimed at determining the effect of hypertonicity due to increased environmental water salinity on gluconeogenesis in air-breathing walking catfish (Clarias batrachus). In situ exposure to hypertonic saline solution (150 mM NaCl) led to a significant stimulation of glucose efflux due to gluconeogenesis from the liver after 7 days with further elevation after 14 days in the presence of each of the three potential gluconeogenic substrates (lactate, pyruvate, and glutamate). This was accompanied by significant increase of activities of three key gluconeogenic enzymes, namely phosphoenolpyruvate carboxykinase (PEPCK), fructose 1,6-biphosphatase (FBPase), and glucose 6-phosphatase (G6Pase) in liver and kidney by about twofold to threefold. Environmental hypertonicity also led to a significant elevation in the levels of PEPCK, FBPase, and G6Pase enzyme proteins in both the tissues by about 2- to 2.75-fold, accompanied by a significant elevation in the level of PEPCK mRNA by about 2- to 2.5-fold after 7 days, and further enhancement to about 3.5- to 4-fold after 14 days. Thus, the upregulation of PEPCK, FBPase. and G6Pase activities appears to be a result of transcriptional regulation of these genes. The induction of gluconeogenesis under environmental hypertonicity, which this catfish faces regularly in its natural habitat, possibly occurs as a consequence of changes in hydration status/cell volume of different cell types. This would certainly assist in maintaining glucose homeostasis, and also for a proper energy supply to support metabolic demands for ion transport and other altered metabolic processes under various environmental hypertonic stress-related insults.
