Hormonal and Nutritional Regulation of IGF-I and Its Binding Proteins

Abstract: The influence of nutrition on growth seems likely to be mediated in part through IGF-I. Restriction of dietary nutrients adversely effects IGF-I synthesis and action at multiple steps, including decreased GH receptors, postreceptor defects in GH action, decreased steady state levels of IGF-I mRNA, and attenuation of IGF-I action. In addition, undernutrition affects the IGF binding proteins in ways that vary from one tissue to another. Understanding the alterations in IGFBPs that are caused by nutritional insuf… Show more

“…It is well known that nutritional status effects IGF-I levels more than IGFBP-3 levels (Baxter and Martin, 1986;Thissen et al, 1994). Low IGF-I levels were observed in chronic disease and malnutrition whereas IGFBP-3 remains relatively unchanged (Baxter and Martin, 1986;Thissen et al, 1994;Underwood et al, 1994). IGFBP-3 correlated with anthropometric measurements in boys but not in girls, suggesting sexual dimorphism.…”

“…Linear growth is directly controlled by GH, in part via IGF-I (26,27), with other factors such as nutrition and the hormonal environment playing modulatory roles (10,28). To the best of our knowledge, the effect of malnutrition on the IGF-I system during puberty has not been reported previously.…”

“…Undernutrition, or poor nutrition, inhibits growth in the laboratory rat (5,10,11) and is one of the major causes of short stature in humans (31,32). In agreement, we now show that undernutrition of peripubertal rats significantly reduced body weight, which at this age is directly proportional to linear growth (33).…”

“…Nutrition is a major regulator of IGF-1 (Underwood, 1996). Fasting results in a decrease while overnutrition results in an increase in IGF-1 levels although the exact nature of the dose -response relationship between food intake and levels of IGFs in circulation remains to be determined.…”
