Homozygous deletions at 3p12 in breast and lung cancer

Abstract: We have constructed a physical map of the region homozygously deleted in the U2020 cell line at 3p12, including the location of putative CpG islands. Adjacent to one of these islands, we have identi®ed and cloned a new gene (DUTT1) and used probes from this gene to detect two other homozygous deletions occurring in lung and breast carcinomas: the smallest deletion is within the gene itself and would result in a truncated protein. The DUTT1 gene is a member of the neural cell adhesion molecule family, although … Show more

“…It is noteworthy that a case of uveal melanomas has been previously described with a translocation at 3p13 as the sole abnormality, suggesting that a second important TSG may reside at this location. 16 Furthermore, DUTT1/ ROBO1 is a putative TSG located within the deleted region identified in the present study, 22,23 the expression of which is absent in uveal melanomas with monosomy 3. 21 DUTT1/ROBO1 is a neural cell adhesion molecule, as is CHL1 at 3p26, and represents a target for further study as a candidate TSG in the preferentially deleted region around 3p12-13.…”

“…In lung tumor cell lines, homozygous deletions have been characterized in regions 3p12, 3p14 and 3p21, for instance in cell line U2020. 116 Interestingly, ROBO1/ Dutt1 was mapped within the deletion and its promoter region is hypermethylated in primary lung, renal and breast tumors. So far, no somatic point mutation of ROBO1 (or of its ligands slits) was reported in tumors.…”

“…High-resolution studies of tumours refined the areas of allelic losses in 3p into several distinct regions representing likely locations of tumour-suppressor genes (TSGs). These candidate regions include 3p12-13, 3p14.2, 3p21.3, 3p24-25 and 3p25-26 (Hung et al, 1995;Sekido et al, 1998;Sundaresan et al, 1998;Wistuba et al, 2000). Investigation of these regions has identified resident TSGs, for example VHL at 3p25 and FHIT at 3p14.2, or likely candidates such as DUTT1/ROBO1 at 3p12.…”
