Homology between mitochondrial DNA of Agaricus bisporus and an internal portion of a linear mitochondrial plasmid of Agaricus bitorquis

Abstract: Agaricus bisporus, the cultivated mushroom, contains a mitochondrial fragment (50H) which was previously demonstrated by Southern hybridization to have sequence similarity to an internal region of pEM, a linear mitochondrial plasmid of Agaricus bitorquis. The nucleotide sequence of 50H was determined and compared to the sequence of the corresponding pEM fragment. The region of sequence homology on pEM is contained within an open reading frame (ORF) that may encode an RNA polymerase, but 50H is neither an intac… Show more

“…2b), we refrained from cloning other fragments of the plasmid. The high AT content of 73% is consistent with data obtained for many other linear plasmids of fungi, such as 74% for the pEM of Agaricus bitorquis (Robison et al, 1991), 67% for the pMC3-2 of Morchella conica (Rohe et al, 1991) and 70% for the kalilo of Neurospora intermedia (Chan et al, 1991).…”

“…We have searched available sequence databases for other examples of split T7-like RNAPs. A number of examples of fragmented, probably inactive enzymes in mitochondrial genomes of fungi (21,51) and Beta vulgaris (7) have been described. Others were found by us in the Spizellomyces punctatus mitochondrial chromosome sequence.…”

“…A reverse transcriptase activity has been proposed to explain the formation and replication of the Mauriceville homology group plasmids (12), while the LaBelle and Fiji plasmids were found to encode a DNA-dependent DNA polymerase (13) like other linear plasmids such as kalilo from Neurospora intermedia (14), maranhar from Neurospora crassa (15), pEM from Agaricus bitorquis (16), pClK1 from Claviceps purpurea (17), pAI2 from Ascobolus immersus (18) and S1 from maize (19).…”
