The quantification of three-dimensional (3D) flow structures and particle dynamics is crucial for unveiling complex interactions in turbulent flows. This review summarizes recent advances in volumetric particle detection and 3D flow velocimetry involving holography. We introduce the fundamental principle of holography and discuss the debilitating depth-of-focus problem, along with methods that have been implemented to circumvent it. The focus of this review is on recent advances in the development of in-line digital holography in general, and digital holographic microscopy in particular. A mathematical background for the numerical reconstruction of digital holograms is followed by a summary of recently introduced 3D particle tracking and velocity measurement techniques. The review concludes with sample applications, including 3D velocity measurements that fully resolve the flow in the inner part of a turbulent boundary layer, the diffusion of oil droplets in high-Reynolds number turbulence, and predator-prey interactions among swimming microorganisms in dense suspensions, as well as oceanic and atmospheric field experiments.

Recent trends in optical metrology suggest that, in order for holographic measurement to become a widespread tool, it must be based on methods that do not require physical development of the hologram. While digital holography has been successfully demonstrated in recent years, unfortunately the limited information capacity of present electronic sensors, such as CCD arrays, is still many orders of magnitude away from directly competing with the high-resolution silver halide plates used in traditional holography. As a result, present digital holographic methods with current electronic sensors cannot record object sizes larger than several hundred microns at high resolution. In this paper, the authors report on the use of bacteriorhodopsin (BR) for digital holography to overcome these limitations. In particular, BR is a real-time recording medium with an information capacity (5000 line-pairs/mm) that even exceeds high resolution photographic film. As such, a centimetre-square area of BR film has the same information capacity of several hundred state-of-the-art CCD cameras. For digital holography, BR temporarily holds the hologram record so that its information content can be digitized for numeric reconstruction. In addition, this paper examines the use of BR for optical reconstruction without chemical development. When correctly managed, it is found that BR is highly effective, in terms of both quality and process time, for three-dimensional holographic measurements. Consequently, several key holographic applications, based on BR, are proposed in this paper.

Laser measurement techniques are becoming more commonly applied to many areas of thermo¯uids and heat transfer. An area of their application, which presents highly challenging requirements, is in the measurement of aerospace¯ows. In this case the experimentalist may encounter high mass¯owrates, high-speed¯ows and in some cases high temperatures, and the measurements may also have to be taken with restricted optical access. Such types of¯ow have already been measured using established laser techniques such as particle image velocimetry, laser Doppler anemometry, laser-induced¯uorescence and liquid crystal thermography. Other more recent techniques which have seen application include Doppler global velocimetry and molecular tagging velocimetry for velocity measurement and thermographic phosphor thermometry for surface temperature measurement. This paper reviews the most recent advances in laser techniques and their application to aerospace environments.
