HO-1 in Bone Biology: Potential Therapeutic Strategies for Osteoporosis

Abstract: Osteoporosis is a prevalent bone disorder characterized by bone mass reduction and deterioration of bone microarchitecture leading to bone fragility and fracture risk. In recent decades, knowledge regarding the etiological mechanisms emphasizes that inflammation, oxidative stress and senescence of bone cells contribute to the development of osteoporosis. Studies have demonstrated that heme oxygenase 1 (HO-1), an inducible enzyme catalyzing heme degradation, exhibits anti-inflammatory, anti-oxidative stress and… Show more

“…This could be explained by the ability of 5-delta aminolevulinic acid to upregulate heme oxygenase 1 (HO-1) as shown in recent studies [33,34]. HO-1 is involved in the normal physiological bone homeostasis, by promoting bone mesenchymal stem cell differentiation, by enhancing osteoblasts functions, and by inhibiting apoptosis and senescence of osteoblasts [35]. In this study, LED irradiation alone did not favor the mineralized nodule formation as well as the 100-ALAD treatment.…”

“…Withdrawal estrogen increased CCR2, CCL7, and CCL2 expression but reduced PMOP in mice after inhibition of CCL7 and CCL2 synthesis by administration of band [ 39 ]. Hmox1 (HO-1) has been identified as a new potential therapeutic target against OP [ 40 , 41 ]. TBX21 gene is a T-box expressed in T cells.…”

“…Much of its effect is linked with the released CO that has widespread interactions with different cells and cell functions. Both HO-1 and HO-2 are identified as therapeutic targets in widely different diseases [ 147 , 148 , 149 , 150 , 151 ]. CO and carbon-monoxide-releasing molecules (CORM) are also investigated as therapeutic potentials [ 152 , 153 ].…”
