HMGA2 Modulates the TGFβ/Smad, TGFβ/ERK and Notch Signaling Pathways in human Lens Epithelial-Mesenchymal Transition.

Abstract: This study indicated that HMGA2 functions as a shared effector in TGFβ2- induced lens fibrosis, modulating the signaling network necessary for EMT in a positive feedback loop.

“…High mobility group AT-hook 2 (HMGA2) was among the identified target genes. According to the literature, HMGA2 mediates epithelial-mesenchymal transition (EMT) and regulates transcription factors linking various signaling pathways, such as the TGF-β and MAPK signaling pathways, to regulators of tumor invasiveness and metastasis (31). miR-18a regulates metastasis in gastric cancer (32), colon cancer (33), breast cancer (34), HCC and nasopharyngeal carcinoma (35,36).…”

“…Meanwhile, after knocking out HMGB1 in mice, the expression of TGF-β is weakened through induction of AGE-BSA dependent RAGE pathway. And the high-mobility group protein 2 (HMGA 2) can be induced by AGEs in HMGA2-knockout mice, which plays a role in inhibiting the formation of reactive oxygen species (ROS) and the activation of p38MAPK (Hou et al 2018). Furthermore, Huang et al (2013) confirmed that neuroglioma associated protein-2 (CLIPR-2) participated in the TIF progress through ERK1/2 signaling pathway in the human proximal tubule cells (HK-2).…”

“…Activation of the PI3K/AKT/mTOR signaling pathway is crucial for EMT processes, which includes cancer cells invasion and migration [166]. Tan et al reported that HMGA2 silencing decreases proliferation of acute myeloid leukemia cells by reducing phosphorylation of AKT and mTOR ( Figure 5D) [93].…”
