HLA-G Gene Polymorphism in Human Placentas: Possible Association of G*0106 Allele with Preeclampsia and Miscarriage

Abstract: Definite causes for several pathologies of pregnancy remain unknown. In light of several recent studies, however, diminished or aberrant HLA-G expression may be associated with certain complication of pregnancy and be linked to HLA-G polymorphism. We analyzed DNA from 60 normal placentas (controls), 140 placentas from miscarriage, 36 placentas from preeclampsia, 76 placentas from fetal hypotrophy, and 34 placentas with hypoxia for variations in coding regions (allelic groups G*0101 to G*0107) and the 14-bp del… Show more

“…The genetic haplotypes associated with these clinical problems are those in which KIR fail to activate uNK cells. [68][69][70] Using explant studies, we recently observed that interactions with trophoblasts stimulate uNK survival and proliferation and decrease uNK cell expression of the activating receptor CD314/NKG2D (Zhang J, Dunk C, Lye JS et al, submitted manuscript). The study of human endometrial and decidual leukocytes is challenging with the global positioning of maternal leukocytes within implantation sites undefined.…”

“…Perhaps HLA-G as an exclusive factor has a very modest effect in relation to risk for RM (Moreau et al, 2008). According to Yan et al (2006), a significantly different distribution of the 14-bp genotype exists among Chinese, Danish and Indian RM populations and the discrepancy may be a result of ethnic variation (see Table 3).…”
