HIV Type 1 Isolate Z321, the Strain Used to Make a Therapeutic HIV Type 1 Immunogen, Is Intersubtype Recombinant

Abstract: The genetic subtypes of human immunodeficiency virus type 1 (HIV-1) display differences in immunologie reactivity1 and possibly transmissibility.2 In addition to the high replication rate of HIV-1 and the infidelity of its reverse transcriptase, interstrain recombination may increase the diversity of phenotypes this virus generates. The discoveries that more than 10% of the HIV-1 strains sequenced to date are intersubtype recombinant3 and that the subtype E viruses prevalent in Thailand are probably subtype A/… Show more

“…delivery and protection against IVAG virus challenge in a mouse model. The HIV-1 Ag used here was an early isolate from an HIV-1-infected individual in Zaire in 1976 (HZ321) that contains a clade A envelope and clade G gag (5,37). Our results show that i.n.…”

“…One of the isoforms identi®ed contained a SH3 domain and a previously undescribed motif, designated the TR domain. The TR domain in Ost was most similar to regions of the Tat protein of HIV (Choi et al, 1997), Ril, a gene downregulated in H-ras-transformed cells (Kiess et al, 1995), and a casein kinase homolog, Rag8, of unknown function. Interestingly, a potential SH3 ligand sequence, (R/K)XXPXXP, present in the TR domain of Ost is conserved in these proteins suggesting that this sequence can also bind SH3 domains.…”

“…For instance, some of the first African HIV-1 isolates to be characterized, MAL and Z321 (obtained from a stored plasma sample obtained in 1976 in a rural area in the northern part of the DRC), have been identified as complex recombinants (8,16,55). The presence of recombinant viruses early in the AIDS epidemic suggests that HIV had been present for a while in this region of Africa.…”
