Historical control data for hematology parameters obtained from toxicity studies performed on different Wistar rat strains: Acceptable value ranges, definition of severity degrees, and vehicle effects

Abstract: The physiological and health status of control animals may vary. Due to this variation, it is important to define acceptable ranges of control hematology parameters to gain a better understanding of adverse and non-adverse effects of test substances. After generating historical control data for two Wistar rat strains (RccHan™:WIST and Crl:WI(Han)) from different breeders, the data sets were statistically analyzed using Minitab®. After noticing that single outliers can affect the study control data set… Show more

“…e blood parameters in the LN18178-treated rats of the main and reversal groups are comparable with the sex-matched control rats, with a few exceptions. e changes (increase or decrease) in parameters are within the limits of normal biological variations depending on the sex and age of the animals [32]. Also, these changes are not dose-related, except the platelet count in the main male rats.…”

“…A confounder was present but could not be identified immediately. After excluding known stratification parameters—such as strain ( Kacew, 1996 ), route of administration ( Gad, 1994 ), sex, age ( Wolford et al, 1987 ; McCutcheon and Marinelli, 2009 ), initial body weight ( Wolford et al, 1987 ), vehicle ( de Kort et al, 2020 ) and the laboratory carrying out the test ( Igl et al, 2019 )—other parameters were checked as possible reasons, such as a change in animal supplier and a potential change in the analytical method. However, none of these parameters provided an explanation for the observed decline in electrolytes in 2017.…”

“…Such changes in experimental animals have a high predictive value for human toxicity (Arika et al, 2016 ). Considering the natural variations of haematological parameters in animals, the adversity of any changes related to BPA treatment have to be evaluated against the background of historical control data (de Kort et al, 2020 ).…”
