Histone H1 Depletion in Mammals Alters Global Chromatin Structure but Causes Specific Changes in Gene Regulation

Abstract: Linker histone H1 plays an important role in chromatin folding in vitro. To study the role of H1 in vivo, mouse embryonic stem cells null for three H1 genes were derived and were found to have 50% of the normal level of H1. H1 depletion caused dramatic chromatin structure changes, including decreased global nucleosome spacing, reduced local chromatin compaction, and decreases in certain core histone modifications. Surprisingly, however, microarray analysis revealed that expression of only a small number of gen… Show more

“…(iii) DNMT3B7 may affect DNA methylation as outlined above, which could in turn lead to alterations in histone modifications. Mice lacking histone H1 showed alterations in DNA methylation levels and consequent gene expression changes in relatively few genes, often in imprinted genes or genes located on the X chromosome (Fan et al, 2005). In addition, histone H3 and H4 acetylation levels increased after patients received 5-azacytidine, a global hypomethylating agent (Gore et al, 2006).…”

“…The participation of different histone H1 subtypes in these processes is not fully elucidated (6). It has been shown that embryonic histone H1 is vital for correct chromosome segregation in Xenopus egg extracts (27).…”

“…The binding of H1 proteins to chromatin is highly dynamic (4,5). Histone H1 stabilizes the nucleosome and is important in compaction of chromatin into higher order structures (6,7). Moreover, it has been implicated in transcriptional regulation (8).…”

“…Several knockout studies across a wide range of organisms revealed that H1 has multiple functions, including participation in the regulation of most nuclear processes (Izzo et al 2008;Happel and Doenecke 2009), mediated at least in some cases by specific H1-protein interactions (McBryant et al 2010). H1 depletion leads to perturbation in the expression of only a subset of genes, with both up-regulation and down-regulation (Shen and Gorovsky 1996;Hellauer et al 2001;Fan et al 2005). This suggests that in vivo, rather than being simply a global repressor, H1 can act as a fine tuner of gene expression.…”
