Histone acetyltransferease p300 modulates TIM4 expression in dendritic cells

Abstract: TIM4 (T cell immunoglobulin mucin domain molecule-4) plays a critical role in the initiation of skewed T helper (Th) 2 polarization. The factors regulating TIM4 expression are unclear. This study tests a hypothesis that p300 and STAT6 (signal transducer and activator transcription-6) regulates TIM4 expression in dendritic cells (DC). In this study, a food allergy mouse model was developed with ovalbumin (a specific antigen) and cholera toxin (CT; an adjuvant). The chromatin immunoprecipitation assay was perfor… Show more

“…Although we found this phenomenon in previous studies, in which we observed that CT increased TIM4 expression to facilitate the development of food allergy in mice1112, while we observed that TIM4 was also associated with the initiation of an allergen-related inflammation in the colon mucosa. We also observed that p300 and STAT6 mediate the microbial product CT-induced TIM4 expression in DCs in a recent study13.…”

“…However, the mechanisms governing Tim-4 expression remain largely elusive. Some evidences suggested that the histone acetyltransferase, p300, and STAT6 regulate the expression of Tim-4 in DCs upon stimulation by the cholera toxin (25). Recently, it was found that cigarette smoke extract (CSE) upregulated Tim-4 expression in immature DCs from murine bone marrow, and the upregulation of Tim-4 stimulated by CSE was inhibited by an ERK inhibitor but not by a p38 or JNK inhibitor (26).…”

“…The data reported above suggest that Gal‐1 may enhance the efficacy of the traditional SIT on allergic responses. To examine this, we developed a food allergy model with OVA and CT (cholera toxin; Sigma‐Aldrich) with the established procedures . From week 5, the sensitized mice were treated with SIT and/or Gal‐1.…”
