Background: Background: As part of a project to evaluate emerging optical technologies for cervical neoplasia, our group is performing quantitative histopathological analyses of biopsy specimens from 1,190 patients. Objectives in the interim analysis are (a) quantitatively assessing progression of the neoplastic process of cervical intraepithelial neoplasia (CIN)/squamous intraepithelial lesions (SIL), (b) detecting malignancy-associated changes (MACs), and (c) phenotypically measuring human papillomavirus (HPV) detected by DNA testing. Methods: The diagnostic region of interest (ROI) from immediately adjacent sections were imaged, and the basal lamina and surface of the superficial layer were delimited. Nonoverlapping quantitatively stained nuclei were selected from 1,190 samples with histopathological characteristics of normal (929), koilocytosis (130), CIN 1 (40), CIN 2 (23), and CIN 3/carcinoma in situ (CIS) (68). A fully automatic procedure located and recorded the center of every nucleus in the region of interest (ROI). We used linear discriminant analysis to assess the changes between normal and CIN 3/CIS.

Purpose: To compare differences in the number of apoptotic bodies and nuclear shapes (size and roundness) between untreated benign prostate hyperplasia (BPH) and untreated advanced prostate adenocarcinoma (PCA) and to elucidate differences in apoptosis number and nuclear shapes with increasing malignant potentiality, by Gleason score, in PCA. Methods: A retrospective study was conducted on 40 patients with BPH and 12 patients with PCA I (Gleason score 2-4), 14 patients with PCA II (Gleason score 5-7) and 14 patients with PCA III (Gleason score 8-10).The frequency of apoptotic bodies (mean percentage calculated from 200 cells/high-power field over 10 fields) was examined on immunostain. Nuclear shapes were determined by an automatic image analyzer. Over 100 hyperplastic cells or cancer cells were detected by the image analyzer. Results:The mean number of apoptotic bodies in BPH and PCA I were not significantly different, but patients with PCA II and PCA III showed significantly higher numbers of apoptotic bodies than patients with BPH. Patients with PCA III had significantly more apoptotic bodies than patients with PCA I. Benign prostate hyperplasia nuclei had the smallest mean nuclear area and the largest mean nuclear peripheral ellipse among the four groups. The study showed that PCA I, II, III nuclei had significantly larger areas and a less circular shape than nuclei from patients with BPH. Nuclei from patients with PCA I were smaller in size and rounder than nuclei from patients with PCA III. Conclusions:The present study clearly shows the presence of apoptosis in BPH and PCA and shows an increasing number of apoptotic bodies with higher cellular malignancy. The nuclear shapes in PCA were more irregular and larger in size with increasing cellular malignancy. As expected, nuclei in BPH were smaller in size and rounder than those of cancer nuclei.

Polymerization of orthopedic cements makes use of a peroxide initiator which is decomposed by an accelerator to provide free radicals. Free radicals which act on the monomer molecules are also known to induce cell lesions and cell death. We used an in vitro model of cement polymerization to study the effects of free radicals release on osteoblast-like cells. Initiation of methylmethacrylate was done with benzoyl peroxide and acceleration by N,N-dimethylaniline. Bulk polymerization was done in calibrated test tubes which were left aging until use. Polymers (aged from J1 to J31 days after completion of the polymerization process) were sawed to produce slices. Slices were rinsed in distilled water and free radical release was measured by spectrophotometric titration with p-iodonitrotetrazolium. Saos-2 osteoblast-like cells were cultured in parallel on the slices. Cells appeared to be round and were altered when grown on slices prepared freshly after polymerization. Cytomorphometric analysis of the cell shape (surface area and form-factor polyethylene confirmed that they spread and flatten on slices prepared a long time after polymerization. Free radical release from polymethylmethacrylate cements is a long-lasting event that can induce bone cells alterations in their neighborhood. Two cytotoxic mechanisms were evidenced: (a) polymer slices released a stable toxic component which could be removed by extensive washing; (b) they released free radicals which were still detectable several days after the end of polymerization. The titration curve was a negative exponential.
