Histological Observation of Islet Hemorrhage Induced by Diagnostic Ultrasound with Contrast Agent in Rat Pancreas

Abstract: Contrast enhanced diagnostic ultrasound CEDUS has been shown to induce capillary hemorrhage in heart and kidney. This study characterized the capillary hemorrhage induced in rat pancreas. The pancreata of anesthetized hairless rats were accessed by laparotomy. A 1.5 MHz diagnostic ultrasound probe with 2.3 MPa peak rarefactional pressure amplitude and 1 s intermittent trigger was used to scan the pancreas, located at the focus (3.8 cm), through saline coupling. The probe was swept to expose the entire organ in… Show more

“…These are manifest primarily as significant hemorrhage in conjunction with vascular disruption (Miller et al, 2011). In the work here, we used a lower pressure of 570 kPa, which caused endothelial cell death and vascular collapse as before (Czarnota et al, 2012).…”

“…The extravascular fibrinogen might bind to cellular antigens, such as intercellular adhesion molecule 1 (ICAM-1) related to early immune responses to injury (Boyd, et al 2008), and would then be labeled immunohistochemically. There were no apparent regions with extensive thrombus formation, such as were seen for contrast enhanced diagnostic ultrasound of rat kidney in Bowman’s capsule (Miller et al 2009) and pancreas around the effected islets (Miller et al 2011a). …”

“…Although clinical trials and meta-analyses have recently dispelled concerns over the safety of UCAs in human patients, [18][19][20][21][22][23] there is a large body of experimental research literature that has not been controverted. Ultrasound and UCAs have induced capillary rupture and hemorrhage, [24][25][26] cardiac damage, 27 and arrhythmias 28 and impaired vascular endothelial function 29 in animal studies. Ultrasound has been shown to mechanically disturb cell membranes, inducing temporary permeability in a phenomenon known as sonoporation.…”
