Summary Introduction: Rosella (Hibiscus sabdariffa L) extract is often used as immune-stimulant because it contains flavonoids, especially anthocyanin and quercetin with antioxidant activities. Objective: This study aimed to determine the safety of the rosella extract consumed in the form of capsules on the vital signs, haematologic parameters as well as kidney and liver function. Methods: This research was conducted using clinical trial pre- and post-test design in healthy participants. There were 21 healthy participants (52% male, age ranged 8–45) consuming rosella capsules for thirty days, in a dose of 500 mg extract daily. Leukocytes, lymphocytes, blood urea nitrogen (BUN), serum glutamic oxaloacetic transaminase (SGOT), serum glutamic pyruvic transaminase (SGPT) and vital signs were consecutively evaluated on days 0, 31, and 45, respectively. The Wilcoxon and paired sample t-test were used to compare the parameters among the evaluated times. Results: The result showed that no significant difference for all parameters among the three time points (p>0.05). Conclusion: These findings suggested that the administration of ethanolic extract of rosella is potential safe and does not negatively affect the vital signs, haemoglobin, leukocytes, lymphocytes, BUN, SGOT, and SGPT.

Introduction: Cadaba farinosa Forsk belongs to the family Cappariraceae used in the treatments of snakebite, breast cancer, and retained placenta in African traditional medicine. However, information about the plant toxicity profile and risk awareness are largely unknown. Aim: To determine the histopathological, biochemical, and hematological effects from aqueous stem bark extract of Cadaba farinosa Forsk on selective internal organs of Wistar rats. Methods: Twenty (20) Wistar rats were randomly divided into five (5) groups of four rats each. The extract was administered by oral gavage in doses of 100, 200, 300, and 400 mg/kg body weight for 28 days. Blood samples were collected for hematological and biochemical analyses, while the liver, kidney, spleen, lung, brain, and stomach were harvested and processed histopathologically using standard methods. Results: Oral administration of the aqueous stem bark extract from Cadaba farinosa Forsk caused a significant increase in hematocrit (P=0.044) and hemoglobin (P=0.046). However, the white blood cell was significantly (P=0.011) reduced at doses of 300 and 400 mg/kg compared to control. The serum albumin of treated animals was also significantly (P=0.017) reduced in a dose-dependent manner compared to control. In addition, histopathological sections of treated animals showed pulmonary hemorrhage, stomach edema, vacuolation in the spleen, and necrosed hepatocytes. Conclusion: Oral administration of the aqueous stem bark extract from Cadaba farinosa Forsk for 28 days is toxic to the liver, spleen, stomach, and lung. Reducing the therapeutic dose and period of exposure may minimize the deleterious effects of plant drugs.

Rosella (Hibiscus sabdariffa L.) calyx traditionally used to treat various diseases. There is limited evidence based could be found including the efficacy and safety. The present study was aimed to investigate the effects of subchronic administration of Rosella calyx extract on the renal function of female Sprague Dawley (SD) rats. The rosella calyx was extracted by 70% ethanol and evaporated with a rotary evaporator to get concentrated extract. The Sprague Dawley rats were randomly divided into five groups including control group which was given CMC Na 0.5% and treated groups were administered orally with the extract of 50, 100, 200 mg/kg bw for 35 days. The satellite group was conducted for observing the reversibility, the observation in this group was conducted 14 days after 35 days of treatment. The biochemical function including ureum and creatinine was analyzed from a blood sample using enzymatic UV-test, and creatinine level were observed by the Jaffe method. The histopathology profile of kidney was observed with Hematoxylin and Eosin staining and followed by microscopic observation. The results showed that ureum levels in treated groups (50, 100, 200, and satellite groups was significantly difference (p<0.05) compare to the control group. The creatinine levels also were found to show significant differences (p<0.05) between the control group with treated groups (50, 100, 200 and satellite group). But the increasing creatinine levels are still within normal range. However, the histopathological profile of animals did not reveal any morphological damage.
