The current study is designed to investigate the histological and immunohistochemical characteristics of the thyroid gland in adult male Sciurus anamalus. This study found that the thyroid gland of the Caucasian squirrel is located in the neck area, below the larynx, on both sides of the trachea. It has two lobes (right and left) with cylindrical shape. The histological studies revealed that the thyroid gland is surrounded by a capsule which consists of connective tissue and forming of two layers which are outer layer and inner layer, and a layer of adipose tissue appears overlapping the outer layer. The inner tissue of the gland consists of follicles with different shapes and sizes, and is lined with simple cuboidal epithelial tissue (follicular cells). Sometimes it appears in the form of high or low cuboidal epithelial tissue, in dependent on the functional activity state of the gland, and the follicles cavities are filled with colloid. In addition, there are two kinds of cells spread in the gland tissue, follicular cells and parafollicular cells, and they are fewer in number and larger in size than follicular cells and appear either singly or in the form of clusters and occupy two sites between the follicles or within the follicle. Immunologically, the results observed an immune response to thyroglobulin antibodies to detect the protein secreted by the follicular cells, which in turn secreted it to the colloid. Moreover, the immune response appeared clear in the cell membrane of the follicular cells and the colloid through the appearance of the brown color which is specific to the immune reaction.

Thyroid ultrasonography examination is widely used in human and small animal medicine. However, it has rarely been applied in cattle. The aim of this study was to determine whether the measurements of the thyroid gland by ultrasound examination correlate to those taken during post-mortem examination. A sample of 22 cows and 23 calves was selected for thyroid gland evaluation. An ultrasound scan was performed ante-mortem, followed by euthanasia (for medical reasons) or slaughtered in the food chain and the dissection of the thyroid gland was therefore performed. Post-mortem, the gland was weighed and its dimensions and volume measured. The volume and weight measurements were compared with the predicted ones on US using the formulas available in the literature. Finally, histological examination was performed on thyroid glands. The dimensions of the thyroid gland measured by ultrasonography were significantly different (p<0.05) from those observed post-mortem, except for lobe lengths in calves (p>0.1). However, in calves, there was no systematic bias between the ultrasound and post-mortem examination of the thyroid gland, which were concordant (with an average error of 18%). Cystic lesions were observed on ultrasound in 9/22 cows and could be found on histological examination in 7 of these. Other lesions, such as follicular hypoplasia and hyperplasia, were seen on histological examination but not on ultrasound. Although the ultrasound measurements did not significantly correlate with those taken post-mortem, this examination may allow to differentiate non-standard thyroids in the case of hyperplastic goiter, as demonstrated in other species. This study also describes and illustrates interesting lesions of the thyroid gland in cattle. These findings are innovative in the description of the use of thyroid ultrasound in cattle, although further studies are needed to allow deeper conclusions.

Heat Stress and Histopathological Changes of Vital Organs: A Novel Approach to Assess Climate Resilience in Farm Animals

Heat stress causes functional and metabolic alterations in different cells and tissues. There are several pathomorphological changes and biomarkers associated with head load in adaptive and productive organs of livestock. Heat stress-induced histopathological alterations in livestock were categorized as degenerative changes (fatty degeneration, steatosis, hydropic degeneration), necrosis (pyknosis, fibrosis), circulatory disturbances (hyperemia, edema, hemorrhage, congestion, thrombosis, ischemia), growth disturbances (hyperplasia, atrophy) and focal/diffuse inflammation (vascular changes, exudation). Upon immunohistochemical analysis, the biomarkers identified in growth-related organs were HSP70, HSP60, GABA, GABAAR, GABABR, HSP90, GnRH, LH, FSH, m6A, Nrf2, and C/EBPβ. The biomarkers in the reproductive organs were HSP70, Bax, Bcl-2, GABA, GABAAR, GABABR, Caspase-3, HSP90, HSPB9, HSPB10, HSF1, HSP40, T, E2, Cyt-C, CAT, BCL2L1, and VEGF. The identified biomarkers in the immune organs were CD3+ T cells, CD4+ T cells, CD8+ T cells, HSP70, and Bcl-2. All these biomarkers could serve as reliable variables in heat stress assessment in livestock. Further, HSP70, HSP90, HSP60, NPY, HSP27, Bcl-2, NF-κB, AQP2, Insulin, CD3+ T cells, CD4+ T cells, CD172a, EGF, AQP1, AQP3, AQP4, AQP5, CRYAB, GHR, 5-HT, CCK, and GLP-1 are heat stress-related biomarkers in adaptive organs that help in assessing the climate resilience of a livestock species and improving understanding about adaptive mechanisms. Among these biomarkers, HSP70 was established to be the ideal cellular biomarker for scaling heat response in livestock. Thus, examining heat-stressed organ histopathology and identifying cellular markers by immunohistochemistry may lay the foundation for screening climate-resilient livestock breeds in the challenging climatic scenario. Further, such an approach could help in developing concepts to combat the detrimental consequences of heat stress to ensure sustainability in livestock production.
